Counting Stomata Biology Harry Introduction: Stomata are tiny holes in a leafs surface essential for the release and exchange of gasses as part of the process of photosynthesis. It is very difficult to look at these directly under a microscope, therefore it is essential to look at an imprint of the stomata. This is a technique. Method: The leaf was placed onto graph paper and its area calculated. This was recorded. A small section of a given leaf was painted with nail polish and then let to dry. A section of clean selo tape was then placed carefully onto the painted section. It was then smoothed over with a pieces of cloth, this removes those pesky little air holes. The tape was peeled of and its was stick onto a slide. Looking at the leaf though the magnifier it was possible to seethe stomata at 10x resolution, however it was too difficult how many there were due...
