David Dawson 13w AS Biology Coursework Aim: To investigate the effect of the substrate concentration, on the rate that it converts hydrogen peroxide to water and oxygen, catalysed by the enzyme peroxidase. Apparatus: * Goggles * Lab Coat * Distilled water * Starting concentration of hydrogen peroxide (labelled 100%) * Yeast suspended in isotonic buffer * Gas prep kits (conical flask with delivery tube and bung) * Gas syringes * Stop watch * Pipettes (1ml, 2ml, 5ml and 10ml) * Pipette fillers Diagram: Method: 1. Collect the apparatus from the apparatus list above and set up as shown in the diagram. 2. Using the following table, measure out 100% concentration of hydrogen peroxide using the 10ml pipette and filler for greater accuracy. Place in the conical flask. Concentration 10% 20% 40% 60% 80% 100% Hydrogen peroxide 2mls 4mls 8mls 12mls 16mls 20mls Water 18mls 16mls 12mls 8mls 4mls 0mls 3. Now measure 10mls of the yeast suspension also using the 10ml pipette and filler. Makes sure you give the yeast a good shake before. Then place in the conical flask and place the bung on straight...
