Mitosis Cell Count in an Onion Root Tip. Aim: To determine the length of time each stage of mitosis lasts in an onion root tip. Hypothesis: I would expect Interphase to take the longest time, as I know that the majority of cells are in this phase at any one time. BY counting all the cells and seeing what stage they are in, I expect that I will be able to work out how long they are in each stage for compared to the other cells. Apparatus: Onion Root Tips. Watch Glasses. Forceps. Slides. Cover Slips. Concentrated HCl (aq). Absolute Ethanol. Glacial Ethanoic/Acetic Acid. Pipette Acetic (Ethanoic) Orcein. A Blunt-Ended Instrument. Blotting Paper Microscope. Method: Place your small cut sections of root tip in a watch glass, which has in it equal amounts of concentrated HCl and absolute ethanol. Wait five to ten minutes. Move the root tips from the watch glass just used to a new watch glass, which has in it 45% glacial ethanoic/acetic acid...
